Electrophysiological Changes of Human-Induced Pluripotent Stem Cell-Derived Cardiomyocytes during Acute Hypoxia and Reoxygenation

Figure 2

Timeline of the experiments. hiPSC-CMs were seeded on 1-wells on glass or MEA 8 days before hypoxia start. Glucose- and serum-free medium was exchanged to the samples on the day before the hypoxia start. 0% O2 hypoxia was induced for 3 hours, after which samples (H) were collected for immunocytochemistry (ICC), western blot (WB), and qPCR, or 19% O2 reoxygenation was induced to the samples. After 24-hour reoxygenation, samples (HR) were collected for ICC, WB, and qPCR. Control samples were kept in a standard incubator for the duration of the hypoxia (CH) or hypoxia-reoxygenation (CHR). For MEA samples, baseline was recorded in 19% O2 after medium exchange, after which 0% O2 hypoxia and 19% O2 reoxygenation were induced as for other samples.
