Insulin Promotes Survival of Amyloid-Beta Oligomers Neuroblastoma Damaged Cells via Caspase 9 Inhibition and Hsp70 Upregulation

Figure 1

Dose dependence of protective effect of insulin against rA 𝛽 42 oligomers induced cell death. (a) LAN5 neuroblastoma cells were untreated (control) or incubated with insulin 50 or 100 or 200  𝜇 M of insulin alone or after incubation with 25 or 40  𝜇 M of rA 𝛽 42. After incubation treated and untreated cells were submitted to viability MTS assay. Cell viability is significantly lower in rA 𝛽 42 25  𝜇 M 𝑃 < . 0 1 , rA 𝛽 42 25  𝜇 M + insulin 50  𝜇 M 𝑃 < . 0 1 versus control, whereas it is not significantly lower with rA 𝛽 42 25  𝜇 M + insulin 100  𝜇 M versus control. Cell viability is significantly lower in rA 𝛽 42 40  𝜇 M 𝑃 < . 0 0 3 , rA 𝛽 42 40  𝜇 M + insulin 50  𝜇 M 𝑃 < . 0 0 3 , whereas it is not significantly lower with A 𝛽 40  𝜇 M + insulin 100  𝜇 M versus control. Percentage of viability is referred to control, the data are the mean ± SD of three separate experiments. (b) Representative morphological images of LAN5 untreated cells (control), treated with rA 𝛽 42 40  𝜇 M, rA 𝛽 42 40  𝜇 M, and insulin 100  𝜇 M, insulin alone 100  𝜇 M. Bar 20  𝜇 m.
