Prolonged Nitric Oxide Exposure Enhances Anoikis Resistance and Migration through Epithelial-Mesenchymal Transition and Caveolin-1 Upregulation

Effect of nitric oxide on epithelial-mesenchymal transition. Cells were exposed to DETA NONOate at various concentrations (10 and 25 μM) for 14 days and subjected to migration and anoikis assays. The expression level of EMT markers including vimentin, snail, TCF8/ZEB1, E-cadherin, ZO-1, N-cadherin, and -catenin were determined using western blot analysis. To confirm equal loading of the samples, the blots were reprobed with -actin antibody. The immunoblot signals were quantified by densitometry. The data are the mean S.D. . versus the nontreated control.
