Effect of NPM on the levels of Bcl-2 family members and caspase 8 activation in Jurkat cells. Jurkat cells were treated with 0.5–1 μM of NPM for 24 h and the cell lysates were analyzed for the levels of antiapoptotic Bcl-2 family proteins (Bcl-2, Bcl-XL, and Mcl-1) (a) and proapoptotic Bcl-2 family proteins (Bad, Bim, Bid, and Bak) (b) by Western blot. To monitor the levels of active cleaved caspase 8, tBID, and BIM over time, Jurkat cells were analyzed by Western blot ((c) and (d)). tBid, truncated Bid; BimEL, Bim extra long isoform; pBimEL, phosphorylated BimEL; tBimEL, truncated BimEL. BimEL is one of the major isoforms produced by alternative splicing. BimEL differs from BimL by the inclusion of exon 3. Actin served as a loading control. Data were from a representative of three independent experiments that gave similar results.
