Colony formation assay. C12-HSL treated (50 μmol/L) and untreated cells were plated in 35 mm cell culture dishes at a density of 2.5 × 10² cells (DU145), 5 × 10² cells (PC3), 5 × 10² cells (LNCaP), and 7 × 10³ cells (RWPE1) per plate. The live cell count numbers were maintained similar in treated and untreated cells. Colonies, defined as groups of ≥20 cells, were identified and designated as a single colony. The plating efficiency was calculated based on number of cells to colony numbers and was about 75%. ; .
