Novel WT1 Target Genes: IL-2, IL-2RB, and IL-2RG Discovered during WT1 Silencing Using Lentiviral-Based RNAi in Myeloid Leukemia Cells

WT1 silencing (WT1-siRNA) induced apoptosis of K562 cells. The K562-WT1-siRNA-GFP⁺ cells (WT1-siRNA) and C-siRNA-GFP⁺ (C-siRNA) control cells were harvested at 0, 3, 6, 12, 24, 48, 72, and 96 hours posttransduction. Flow cytometry analysis was performed based on Annexin V-FITC/PI staining for apoptotic cell determination. Early apoptotic (a) and late apoptotic (b) cells were calculated based on the percentage of Annexin V⁺ and Annexin V⁺/PI⁺ cell population, respectively. The assay was performed in triplicate ( by paired samples -test).
