In Vivo Suppression of Autophagy via Lentiviral shRNA Targeting Atg5 Improves Lupus-Like Syndrome
Figure 2
shRNA targeting on Atg5 may efficiently suppress gene expression and autophagy in vitro. (a) Mouse lung cancer cell line, LLC-1, was infected with shLuc- or shAtg5-containing lentivirus. Atg5 expression levels were measured by RT-qPCR (Left) and western blot (Right). The numbers show the relative expression of Atg5. (b) Virus-infected LLC-1 cells were starved for 12 and 24 hours. The cells were then stained with AO and analyzed by flow cytometry to determine the autophagy levels (AVOs (%)). The results are representative of three independent experiments. (c, d) The level of LC3 in LLC-1 cells was evaluated using western blot after 24 hours of starvation or Rapamycin treatment (100 nM). The numbers show the relative expression of the LC3-II/I ratio in the individual group. Mean values were shown by a bar. represents .
