IL-6/STAT3 increased the expression of the proteasome 20S α+β. IL-6 (30 pg/mL) or PBS was used to activate HCT116 cells, taking samples at the indicated time. (a) Cells were fixed and stained as described: proteasome 20S α+β (green) and DAPI (blue). (b) Proteasome 20S α+β and GADPH immunoblots were generated from lysates. (c and d) For 12 hours, HCT116 cells were stimulated with either IL-6 (30 pg/mL). qRT-PCR was used to determine the proteasome 20S α (PSMA1-A7) and proteasome 20S β (PSMB1-B7) mRNA levels.
