Overcoming Basal Autophagy, Kangai Injection Enhances Cisplatin Cytotoxicity by Regulating FOXO3a-Dependent Autophagic Cell Death and Apoptosis in Human Lung Adenocarcinoma A549/DDP Cells

A549/DDP cells showed a higher level of basal autophagy in response to cisplatin treatment. (a) Direct cytotoxic effects of cisplatin in A549 and A549/DDP cells. A549 and A549/DDP cells were treated with various concentrations (0, 1, 2, 4, 8, 16, 32, 64, and 128 μM) of cisplatin for 24 h. (b) RNA sequencing was used to compare the differentially expressed genes between A549 and A549/DDP cells. (c) Western blot analysis revealed the relative expression levels of autophagy-related proteins (Beclin 1, LC3 II, and p62) in A549 cells and A549/DDP cells. Every experiment result was repeated ≥3 times, and results are presented using values ().
