Overcoming Basal Autophagy, Kangai Injection Enhances Cisplatin Cytotoxicity by Regulating FOXO3a-Dependent Autophagic Cell Death and Apoptosis in Human Lung Adenocarcinoma A549/DDP Cells

<div>A549/DDP cells showed a higher level of basal autophagy in response to cisplatin treatment. (a) Direct cytotoxic effects of cisplatin in A549 and A549/DDP cells. A549 and A549/DDP cells were treated with various concentrations (0, 1, 2, 4, 8, 16, 32, 64, and 128 <i>μ</i>M) of cisplatin for 24 h. (b) RNA sequencing was used to compare the differentially expressed genes between A549 and A549/DDP cells. (c) Western blot analysis revealed the relative expression levels of autophagy-related proteins (Beclin 1, LC3 II, and p62) in A549 cells and A549/DDP cells.
