CRISPR/Cas9-mediated NHEJ and HDR. When DSBs occur, cells will use the NHEJ or HDR mechanism for DNA repair. The NHEJ mechanism will directly link the broken DNA together and can be utilized for gene knockout. The HDR mechanism repairs the broken DNA by homologous recombination and can be utilized for gene knock-in [37].
