Increased Proportion of Dual-Positive Th2–Th17 Cells Promotes a More Severe Subtype of Asthma

Dual-positive Th2–Th17 cells promote further differentiation and function of T cells into the same biphenotypic cells. Mice received dual-positive Th2–Th17 cells obtained from asthma/differentiation mice (adoption/asthma group) or naïve CD4+ T cells (asthma group) separately and then received allergens to build an asthma model. Twenty-four hours after the last challenge, spleen CD4+ T cells and blood serum were harvested for various studies. (a) The proportion of dual-positive Th2–Th17 cells was assessed by flow cytometry. (b) mRNA expression of GATA3 and RORγt in spleen CD4+ T cells was detected by quantitative real-time PCR. (c) Secreted IL-4 and IL-17 in supernatants were evaluated using ELISA. .
