Identification of IL-38 and IL-36 overexpression construct. (a) SW982 cells were transfected with EGFP fusion IL-36 or IL-38 encoding plasmids or empty vector. Cells were examined under microscopy (100x) for EGFP expression. Representative images are presented from three biological replicates. (b) SW982 cells were transfected with empty vector or IL-36 (left panel) or IL-38 (right panel) or left untransfected, followed by total RNA isolation 24 hours posttransfection and subjected to qRT-PCR analysis. (c) SW982 cells were transfected with the indicated plasmids for 24 hours followed by Western blot detection for IL-36 or IL-38 expression; GAPDH was used as control.
