Chlorin e6 Prevents ADP-Induced Platelet Aggregation by Decreasing PI3K-Akt Phosphorylation and Promoting cAMP Production

Effects of CE6 on fibrinogen binding to integrin in ADP-activated platelets. The inhibitory effects of CE6 on fibrinogen binding to integrin in ADP-stimulated platelets were measured by flow cytometry (a). Washed platelets ( /mL) were pretreated with vehicle (DMSO) or CE6 at concentrations ranging from 3.25 μM to 30 μM. ADP (10 μM) was then incubated with human fibrinogen labeled with Alexa Fluor 488 (20 μg/mL) for 5 min. The cells were subsequently fixed with 0.5% paraformaldehyde at 4°C for 30 min. Graphs showing fluorescent intensity present the data from one experiment but are representative of four independent trials. Data are expressed as the mean fluorescence intensity (MFI) of fibrinogen-positive platelets. Each graph presents the results expressed as percent of gated (A). and compared to the agonist control.
