[6]-Gingerol Prevents Disassembly of Cell Junctions and Activities of MMPs in Invasive Human Pancreas Cancer Cells through ERK/NF-κB/Snail Signal Transduction Pathway

Effect of [6]-gingerol on the DNA-binding activity of NF-κB in PANC-1 cells. (a) Nuclear and cytosolic extracts were prepared using the NE-PER nuclear and cytoplasmic extraction kit from PANC-1 cells treated with [6]-gingerol for 24 h. Western blotting was then performed on each fraction. Actin was used as an internal control for, the cytosolic fraction, and lamin was applied as an internal control for the nuclear fraction. (b) Cells cultured under the same conditions were pretreated for 30 min with U0126, an ERK inhibitor. Cells were then tested for the DNA-binding activity of NF-κB by EMSA. Band intensities in the immunoblots were quantified by densitometry using L Process and MultiGauge software. Band intensities were normalized relative to the internal control and background. Data represent the mean ± SD from three independent experiments. Statistical analyses (Student's -test and one-way ANOVA) were performed using GraphPad Prism 5. * versus the untreated control. + , [6]-gingerol compared with U0126-treated cells.
