DSGOST inhibition of cold-induced RhoA activation in endothelial cells. (a) HUVEC or HDMEC were pretreated with DSGOST at different concentrations (50, 100, 150, and 200 μg/mL) for 30 minutes, exposed to 25°C for another 30 minutes, and then subjected to GST pull-down assays for RhoA activity. (b) Cells were transfected with SRE-luc construct for 48 hours, pretreated with DSGOST (100 μg/mL) for 30 minutes, and then exposed to cold (25°C) for another 90 minutes, . (c) Cells were transfected with CA-RhoA and SRE-luc constructs for 48 hours, treated with DSGOST (100 μg/mL) for 2 hours, and then subjected to the reporter assays. Experiments were performed in triplicate and repeated three times independently. Data represent mean ± SEM.
