Effects of incubation time and microsome concentration on the metabolism of prednisone in rat liver and kidney microsomes. (a) The incubation system containing 5 μg/μL of the liver microsomes was incubated for 5, 10, 20, 25, 30, or 45 min. The analyte was injected into HPLC system to determine the effect of incubation time on the metabolism of prednisone in rat liver microsomes. (b) The incubation system containing different concentration of liver microsomes (0.5~5 μg/μL) was incubated for 25 min. The analyte was injected into HPLC system to determine the effect of microsome concentration on the metabolism of prednisone in rat liver microsomes. (c) The incubation system containing 3 μg/μL of the kidney microsomes was incubated for 5, 10, 20, 25, 30, or 45 min. The analyte was injected into HPLC system to determine the effect of incubation time on the metabolism of prednisolone in rat kidney microsomes. (d) The incubation system containing different concentration of kidney microsomes (0.5~5 μg/μL) was incubated for 45 min.
