Hydroxysafflor Yellow A Inhibits TNF-α-Induced Inflammation of Human Fetal Lung Fibroblasts via NF-κB Signaling Pathway

Figure 4

HSYA inhibits TNF-α induced proliferation and inflammatory response in MRC-5 cells via the NF-κB/AP-1 signaling pathway. (a) Western blot was used to detect the level of IKK and p65 phosphorylation and protein expression level of IκBα; (b–d) quantitative evaluation on phosphorylated levels of IKK, p65, and IκBα protein expressions; (e) luciferase reporter assay was used to detect the fluorescent activity of TGF-β1-TA. Chromatin immunoprecipitation was used to detect the binding effect of AP-1 and TGF-β1-TA using anti-AP-1 as antibody in immunoprecipitation; (f) the expression of TGF-β1-TA was detected by PCR; (g) TGF-β1-TA band was detected by agarose gel electrophoresis; HSYA, hydroxysafflor yellow A; ns, ; ; .
