Sesamin Promotes Neurite Outgrowth under Insufficient Nerve Growth Factor Condition in PC12 Cells through ERK1/2 Pathway and SIRT1 Modulation

To confirm the correlation of SIRT1 effect and neuronal differentiation, cells were treated with JGB1741, the selective SIRT1 inhibitor under combination treatment. (a) Cell lysates were collected at 4 h to investigate the ERK signaling molecules, (b) while at 72 h for the synaptophysin and GAP43 protein measurement by western blot analysis. Band intensities are shown as the bar graph. Moreover, (c) the PC12 morphology was observed after 72 h by using an inverted phase-contrast microscope. (d) The percentage of differentiation and (e) neurite length were calculated. Data represent the means ± SD of three independent experiments. and compared with the NGF alone-treated group; and compared with the combination treatment group.
