Sesamin Promotes Neurite Outgrowth under Insufficient Nerve Growth Factor Condition in PC12 Cells through ERK1/2 Pathway and SIRT1 Modulation

<div>To confirm the correlation of SIRT1 effect and neuronal differentiation, cells were treated with JGB1741, the selective SIRT1 inhibitor under combination treatment. (a) Cell lysates were collected at 4 h to investigate the ERK signaling molecules, (b) while at 72 h for the synaptophysin and GAP43 protein measurement by western blot analysis. Band intensities are shown as the bar graph. Moreover, (c) the PC12 morphology was observed after 72 h by using an inverted phase-contrast microscope. (d) The percentage of differentiation and (e) neurite length were calculated.
