Constitutive Androstane Receptor-Mediated Inhibition of Metformin on Phase II Metabolic Enzyme SULT2A1
Figure 2
Metformin represses CITCO-induced SULT2A1 expression. (a–c) HepaRG cells were treated with RIF (20 μM), CITCO (1 μM), metformin (0.1, 0.5, 1 mM), or their combination for 24 h. Cells treated only with DMSO were considered as the control group. Total mRNA was collected and the expressions of SULT2A1 and β-actin were analyzed using real-time PCR. Values were normalized to the expression of β-actin. (d–f) HepaRG cells were treated with RIF (20 μM), CITCO (1 μM), metformin (0.1, 0.5, 1 mM), or their combination for 48 h. Whole-cell extracts were harvested, and the expression of SULT2A1 and the internal control β-actin were analyzed using western blotting. Quantitation of the SULT2A1 protein bands was normalized to β-actin. A representative blot was shown and quantified by Image J software. The data shown are the mean ± SEM (n = 3) ( and , comparison indicated by brackets. ns: not significant).
