Hydroxyurea-Induced Replication Stress

BLM-downregulated HeLa cells and BS cells are insensitive to HU-induced replication stress. (a) HeLaV and HeLashBLM cells were transfected with 100 nM of a negative control siRNA (ctrl) or with 100 nM of siRNA targeting the BLM mRNA (BLM), respectively. After 24 h, cells were left untreated or were treated with 2 mM or 5 mM HU for 16 h. Cells were then plated in triplicate at three densities in drug-free medium. The means of six independent experiments for 2 mM HU and of two independent experiments for 5 mM HU are shown. The error bars indicate SD. (b) As in (a) except that cells were treated with 2 mM HU for 48 hr, the means of 3 independent experiments are shown.
