Exosomal MiR-1290 Promotes Angiogenesis of Hepatocellular Carcinoma via Targeting SMEK1

Figure 2

Exosomal miR-1290 targets endothelial cells and exerts an angiogenic effect in vitro. (a) The expression level of miR-1290 in HUVECs, HepG2, SMMC-7721, and L-02 cells were detected by qRT-PCR. (b) Exosomes were isolated from the conditioned medium of DiI-labeled (DiI+) or control (DiI-) HCCs and applied to culture with DiO-labeled HUVECs for 24 (h) followed by confocal microscope detection. (c) Exosomes were isolated from the conditioned medium of SMMC-7721 cells that transfected with miR-1290-FAM or miR-1290 mimic and applied to culture HUVECs for 6 h before detection for fluorescent FAM. (d) Cell viabilities of SMMC-7721 cellstransfected with 50 nM miR-1290 mimic, miR-1290 inhibitor, or the negative control were detected by CCK-8 assay. (e, f, g) The tube formation (e), wound-healing (f), and Transwell (g) assays of HUVECs were performed in HUVECs that transfected with 50 nM miR-1290 mimic, miR-1290 inhibitor, or the negative control. ; ; ; .
