Attenuation of -Amyloid-Induced Oxidative Cell Death by Sulforaphane via Activation of NF-E2-Related Factor 2

Figure 2

Protective effect of SUL on A -induced pro-apoptotic signals. SH-SY5Y cells were exposed to 15  M of A   in the presence or absence of SUL (2  M and 5  M) for 24 h. Activation of JNK (a) and expression of Bcl-2 family proteins (b) were assessed by western blot analysis using anti-phospho-JNK, anti-JNK, anti-Bcl-2, anti-Bax, and anti-actin antibodies. Actin levels were monitored to verify equal amount of protein loading. Relative expression levels of p-JNK/JNK and Bax/Bcl-2 were quantified from three independent experiments and are represented on the right panels. (c) Mitochondria membrane potential was measured by immunofluorescence staining using TMRE probe. The representative images of TMRE fluorescence were shown. (A) Vehicle-treated control; (B) A alone (15  M); (C) A (15  M) + SUL (5  M).
