Blockage of NOX2/MAPK/NF-κB Pathway Protects Photoreceptors against Glucose Deprivation-Induced Cell Death

Inhibiting MAPK signal protects 661W cells from GD-induced death. Cell death was evaluated with PI/Hoechst staining. PI-positive cells were identified with purple fluorescence, and total cells were counter stained with Hoechst showing blue fluorescent nuclei. (a) 661W cells were cultured in glucose-free medium for 16 h as a vehicle control. 661W cells were treated with U0126 (1–50 μM), SP600125 (10-100 μM), SB203580 (10-100 μM), and PD98059 (1–50 μM) during GD. Scale bar = 200 μm. (b) Quantitative assessment of cell death was performed through PI/Hoechst staining. Scale bar = 200 μm. Assays were performed in quadruplicate, and data are shown as means ± SEM ( compared to the vehicle group; one-way ANOVA and Bonferroni test).
