The Synthetic Lignan Secoisolariciresinol Diglucoside Prevents Asbestos-Induced NLRP3 Inflammasome Activation in Murine Macrophages

LGM2605 inhibits NF-κB expression and prevents asbestos-induced iNOS expression and nitric oxide production by murine peritoneal macrophages. The concentrations (μM) of nitrates and nitrites (a) were determined in cell culture medium at 0, 1, 2, 4, 6, 8, 12, and 24 hours post asbestos exposure. Macrophage mRNA expression iNOS (b) was determined at 0, 8, and 24 hours post asbestos exposure using qPCR and protein levels of iNOS were evaluated by Western blotting for iNOS (c) (molecular weight 130 kDa). Levels of active nuclear NF-κB (d) were determined at 0, 1, 2, 4, 6, 8, and 12 hours post asbestos exposure, while mRNA expression of NF-κB (e) was determined at 0, 8, and 24 hours post asbestos exposure using qPCR. Levels of target gene mRNA were normalized to β-actin RNA, and values are expressed as mean fold change from CTL at time 0.
