Camalexin Induces Apoptosis via the ROS-ER Stress-Mitochondrial Apoptosis Pathway in AML Cells

Camalexin induces ROS generation in leukemia cells. (a) NB4 and HL-60 cells were treated with various doses of camalexin for 12 h, and then intracellular ROS was measured by flow cytometry. (b) NB4 and HL-60 cells were pretreated with or without NAC (20 μM) for 4 h, and then cells were incubated with camalexin (40 μM) for 24 h and cellular apoptosis was measured by flow cytometry. (c) NB4 and HL-60 cells were pretreated with or without NAC (20 μM) for 4 h, and then cells were incubated with camalexin (40 μM) for 48 h and disruption of MMP was measured. (d, e) NB4 and HL-60 cells were pretreated with or without NAC (20 μM) for 4 h, and then cells were incubated with camalexin (40 μM) for 24 h and total cellular lysates were subjected to Western blot analysis with indicated antibodies. (f, g) NB4 and HL-60 cells were treated as described above, and then the release of mitochondrial proteins and activation of Bax were assayed by Western blotting.
