The effect of melatonin on cell proliferation and matrix synthesis in human articular chondrocytes. (a) Chondrocytes were treated with melatonin (MT) at concentrations of 1 μM and 100 μM, and cell proliferation was evaluated using the CCK-8 assay. Untreated cells served as the control group (CTRL) and cells treated with ethanol (EtOH) served as the vehicle group. (b–d) The mRNA levels of chondrogenic genes, including COL2A1 (b), ACAN (c), and SOX9 (d), were quantified with real-time PCR using GAPDH for normalization. (e) The protein expression of type II collagen was confirmed through immunofluorescence. . Values are the of six independent experiments () in cell proliferation assays and four independent experiments () in PCR experiments. Statistically significant differences are indicated by where or where between the indicated groups.
