Downregulation of the Proton-Activated Cl- Channel TMEM206 Inhibits Malignant Properties of Human Osteosarcoma Cells
Figure 7
Effects of silencing TMEM206 on OS metastasis in vivo. (a, b) Silencing TMEM206 inhibited the tumor growth in the xenograft nude mice. Nude mice were subcutaneously injected with MG63 cells transfected with TMEM206-shRNA by lentivirus infection. Tumor sizes were measured every 7 days, and the mice were sacrificed on day 21, and the tumors were excised for analysis (, , as comparing with SC-shRNA group, respectively). (c) Representative immunohistochemical staining with TMEM206 and β-catenin antibody of the xenograft tumor. Scale bar: 50 μm. (d) Representative images of whole lungs from nude mice inoculated with MG63 cells and the statistical analysis showed the metastatic foci of the lung (, as comparing with SC-shRNA group, respectively). Black arrowhead represent the metastatic foci in the lung. (e) Schematic diagram of the mechanism that TMEM206 regulating OS. The data are represented as from 3 replicate experiments.
