[Retracted] Stearoyl-CoA Desaturase 1 Potentiates Hypoxic plus Nutrient-Deprived Pancreatic Cancer Cell Ferroptosis Resistance
Figure 1
H/NS condition protects pancreatic cancer cells from ferroptosis. (a) PANC1 and Patu8988 cells were treated with erastin (5 μM for PANC1 and 40 μM for Patu8988), RSL3 (3 μM for PANC1 or 5 μM for Patu8988), or sulfasalazine (2 mM) cultured in normal or H/NS condition for 24 hours. Cell viability was measured using a CCK8 kit. (b) Representative images showing the induction of cell death in PANC1 cells treated with erastin (5 μM), RSL3 (3 μM), or sulfasalazine (2 mM) cultured under normal or H/NS condition with or without Fer-1 for 24 hours. (c) Cell viability was measured in Ctrl shRNA and GPX4 shRNA PANC1 cells cultured in normal or H/NS condition for 24 h. (d) PANC1 and Patu8988 cells were treated with DMSO (control), erastin (5 μM for PANC1 and 40 μM for Patu8988), RSL3 (3 μM for PANC1 or 5 μM for Patu8988), and sulfasalazine (2 mM) in normal or H/NS condition for 24 hours. The levels of MDA were assayed. SAS represents for sulfasalazine; RSL3 represents for (1S,3R)-RSL3; Fer-1 represents for ferrostatin-1; GPX4 represents for glutathione peroxidase 4; Nor represents for normal condition. H/NS represents for hypoxia combination with nutrient starvation condition. Experiments were repeated three times and the data are expressed as the . . . . .
