Four-Octyl Itaconate Protects Chondrocytes against H2O2-Induced Oxidative Injury and Attenuates Osteoarthritis Progression by Activating Nrf2 Signaling

Keap1 knockout abolishes OI-induced cytoprotection from H2O2. Stable chondrocytes with the CRISPR/Cas9-Keap1-KO-GFP construct (ko-Keap1) or the CRISPR/Cas9 control vector (“Cas9-C”) were established, and the expression of the listed genes was tested (a, b). Following treatment with OI (25 μM), chondrocytes were added by H2O2 (300 μM). Cell viability and apoptosis were assessed using CCK-8 (c) and Trypan blue assays (d), respectively.
