OST inhibited RANKL-induced osteoclast formation in vitro. (a) Representative images of TRAP-stained multinucleated osteoclasts treated with OST. (b, c) The number and size (in terms of area) of TRAP-positive multinucleated osteoclasts with three or more nuclei were quantified. (d) OST suppressed RANKL-induced expression of the osteoclast marker genes. Real-time quantitative PCR was conducted using RNA extracted from the BMM-derived osteoclasts treated with the indicated concentrations of OST for 5 days. The expression of NFATc1, c-Fos, DC-STAMP, V-ATPase d2, TRAP, CTR, and CTSK was normalized to the expression of the housekeeping gene GAPDH. (, , ).
