Cryptochrome and Period Proteins Are Regulated by the CLOCK/BMAL1 Gene: Crosstalk between the PPARs/RXR 𝛼 -Regulated and CLOCK/BMAL1-Regulated Systems

The effect of the CLOCK/BMAL1 heterodimer on PPAR target genes. Dose-dependent changes in activity of the 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) synthase promoter (a) induced by the CLOCK/BMAL1 gene (0–140 ng) in the presence and absence of fenofibric acid (10 M) in CV-1 monkey kidney cells plus the PPAR /RXR gene (50 ng). When the promoter of the HMG-CoA synthase gene did not contain direct repeat sequences (del-pHMG-Luc), no change in transcriptional activity of the HMG-CoA synthase promoter was observed (b). The luciferase activity of pHMG-Luc was normalized to the luciferase activity of pRL-TK. Experiments were performed in triplicate, and four independent experiments were performed. All data are means ± SD.
