The Nitrated Fatty Acid 10-Nitro-oleate Diminishes Severity of LPS-Induced Acute Lung Injury in Mice

OA-NO₂ reduces inflammatory phenotype in LPS-activated alveolar macrophages. Induction of ALI by i.t. injection of LPS (50 μg) was followed 30 min later by i.t. administration (50 μL) of OA-NO₂ (50 μg) or vehicle (10% DMSO). After another 5.5 h BAL fluid was obtained. Alveolar macrophages were isolated from the BAL fluid and plated in DMEM + 10% FBS. After 1 h, RNA was isolated and expression of the indicated genes was determined using real-time PCR; results were normalized to values for the housekeeping genes glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and 9s rRNA. Data are representative of one of two independent experiments with 𝑛=6–8 mice per group; *𝑃<0.05; **𝑃<0.01; ***𝑃<0.001.
