Oncostatin M and TLR-4 Ligand Synergize to Induce MCP-1, IL-6, and VEGF in Human Aortic Adventitial Fibroblasts and Smooth Muscle Cells

OSM and LPS regulation of MCP-1, IL-6, VEGF, and IL-8 expression in HAoSMC. HAoSMCs were cultured as in Section 2 and then stimulated with increasing doses of LPS alone or with 0.5 or 5 ng/mL OSM as indicated ((a), (c), (e), and (g)). In ((b), (d), (f), and (h)), HAoSMCs were stimulated with 5 ng/mL gp130 cytokine (OSM, LIF, IL-31, IL-11, and IL-6), alone or in combination with 10 ng/mL LPS as indicated. Supernatants were collected after 18 hours, frozen, and later analyzed by ELISA for MCP-1 ((a), (b)), IL-6 ((c), (d)), VEGF ((e), (f)), and IL-8 ((g), (h)). Results are representative of multiple separate experiments. Statistical significance: ^∗𝑃≤0.05, ^∗∗𝑃≤0.01, compared to LPS alone at the same concentration.
