CD137 Regulates NFATc1 Expression in Mouse VSMCs through TRAF6/NF-κB p65 Signaling Pathway

Silencing the expression of TRAF6 significantly attenuated the agonist-CD137mAb induced increasing of NF-κB p65 and NFATc1 in VSMCs. The VSMCs were treated with agonist-CD137mAb (10 mg/L) for 12 h after transfection of siTRAF6. Total RNA and cytoplasm and nucleus protein were isolated from the harvested cells. The cells were harvested for examination of mRNA levels of NFATc1 (a) by qRT-PCR and protein levels of NFATc1 (b) and p-p65 cytoplasm (c) and nucleus (d) by Western blot.
