HT-29 and Caco-2 Reporter Cell Lines for Functional Studies of Nuclear Factor Kappa B Activation

NF-κB activation time course kinetics for different clones. Caco-2-NF-κB-hrGFP C3 (black bar) and D5 (grey bar) clones (a) were seeded at 1 × 10⁵ cell/well, and HT-29-NF-κB-hrGFP E5 (black bar) and F6 (grey bar) clones (b) were seeded at 5 × 10⁴ cell/well. Cells were cultured ON and treated with 50 ng/mL of TNF-α over time. Cells were trypsinized and GFP expression was analyzed at different time points by flow cytometry. Data was expressed as mean of triplicates with SD error bars. NF-κB activation was determined by the subtraction of the % GFP⁺ of nonstimulated cells from the % GFP⁺ of stimulated cells.
