Multiple Immunosuppressive Effects of CpG-c41 on Intracellular TLR-Mediated Inflammation

CpG-c41 affects inflammasome formation and activation. RAW264.7 cells were seeded into 6-well culture plates at 3 × 10⁶/ml/well and BMDMs were seeded into 96-well culture plates at 5 × 10⁵/200 μl/well. Cells were stimulated with polyI:C (100 μg/ml), LPS (100 ng/ml), R848 (1 μg/ml), or CpG-1826 (3 μM) in the presence or absence of CpG-c41 (8 μM) for 4 hours. Cells were then stimulated with ATP (5 mM) for 30 min. (a) NLRP3 and caspase-1 protein expression in RAW264.7 cells were detected by WB, with tubulin as an internal control.
