Involvement of Oxidative Stress and the Epidermal Growth Factor Receptor in Diesel Exhaust Particle-Induced Expression of Inflammatory Mediators in Human Mononuclear Cells

Oxidative stress is required for DEP-induced expression of IL-8 and TNFα in THP-1 cells. (a) THP-1 cells were treated with 10-100 μg/ml DEP for 4 h, and intracellular ROS levels were measured using the fluorescent probe carboxy-H2 DCFDA and flow cytometer. (b) THP-1 cells were pretreated with 10 mM NAC for 2 h, prior to DEP (100 μg/ml) treatment for another 4 h. ROS was detected as described previously. Furthermore, THP-1 cells were pretreated with 10 mM NAC for 2 h, prior to DEP treatment for another 24 h.
