lncRNA-SNHG14 Plays a Role in Acute Lung Injury Induced by Lipopolysaccharide through Regulating Autophagy via miR-223-3p/Foxo3a

Figure 4

lncRNA-SNHG14/miR-223-3p/Foxo3a axis participated in LPS-induced A549 cells injury. A549 cells were transfected with pcDNA3.1-SNHG14, pcDNA3.1-SNHG14 + siRNA-Foxo3a, miR-223-3p, and miR-223-3p + siRNA-Foxo3a, respectively. Cell viability was detected using CCK-8 kit at 0, 24, 48, and 72 h (a, b). The apoptosis level of cells in each group was measured using RealTime-Glo™ Annexin V dye. Luminescence was detected using a fluorescence plate reader to indicate apoptosis (c, d). Cells were transfected with ASO-SNHG14, ASO-SNHG14 + pcDNA3.1-Foxo3a, miR-223-3p mimic, and miR-223-3p mimic+pcDNA3.1-Foxo3a, respectively. Cell viability (e, f), apoptotic cells (g, h), and cytokine level changes (i, j) were detected under LPS treating condition. compared with transfection negative control mix or LPS untreated control. # compared with cells without cotransfection of siRNA-Foxo3 or pcDNA3.1-Foxo3a.
