Exosomes Containing LINC00636 Inhibit MAPK1 through the miR-450a-2-3p Overexpression in Human Pericardial Fluid and Improve Cardiac Fibrosis in Patients with Atrial Fibrillation

miR-450a-2-3p decreases TGF-β-induced activation and proliferation of CFs. miR-450a-2-3p, α-SMA, COL1, and COL3 mRNA expression in CFs transfected with mimics (a)–(d) and inhibitor (i)–(l) was detected using RT-qPCR. The protein levels of α-SMA, COL1, and COL3 in CFs transfected with mimics (e)–(h) and inhibitor (m)–(p) were determined using western blotting. The expression level of the miRNAs analyzed was normalized to the expression level of U6, and the expression level of mRNAs and proteins was normalized to the expression level of β-actin. Representative images of immunofluorescence staining for α-SMA in CFs transfected with mimics and inhibitor (r) (200× magnification) showed the antifibrotic effect of miR-450a-2-3p.
