Influence of Inflammatory Cytokines IL-1β and IFNγ on Sarcoplasmic Aggregation of p62 and TDP-43 in Myotubes

Figure 6

p62 expression was increased with IL-1β and LC3II/LC3I ratio increased with IL-1β + IFNγ treatment. (a) Western blot of p62, TDP-43, and LC3 with GAPDH loading control. (b) p62 densitometry normalized to GAPDH. There was an increase compared to control for p62 protein expression with IL-1β treatment (), but no difference between control and the other cytokine treatments. (c) Densitometry analysis of TDP-43 protein expression normalized to GAPDH showed no difference in TDP-43 expression between any of the conditions. (d) Ratio of LC3II to LC3I expression. IL-1β + IFNγ increased LC3II/LC3I ratio compared to the control (). There was no difference compared to the control for IL-1β, or IFNγ. (e) Total LC3II relative to GAPDH. There was no difference between any of the conditions (one-way ANOVA ). One-way ANOVA with Dunnett’s multiple comparisons. n = 6 myogenic donors.
