miR-100-5p Promotes Epidermal Stem Cell Proliferation through Targeting MTMR3 to Activate PIP3/AKT and ERK Signaling Pathways

miR-100-5p promotes EpSC proliferation in the organotypic skin through MTMR3. (a) Human EpSCs were cultured with 20 μg ADSC-Exos (Exos), miR-100-5p-enriched ADSC-Exos (miR-100-Exos), or same volume of PBS for different periods of time and examined for cell proliferation by MTT assay. (b–d) Human skin explants intradermally injected with PBS, Exos, miR-100-Exos, or miR-100-Exos + MTMR3 expression plasmid (MTMR3) were cultured for 2 and 5 days. Histology was examined by HE staining; cell proliferation and EpSCs were examined by immunohistochemical staining of PCNA, β1 integrin and α6 integrin. miR-100-5p and MTMR3 mRNA levels were examined by RT-qPCR.
